Dionex Demonstrates Simplified Sample Preparation Method for HPAE-PAD Analysis

Dionex announces a new method that simplifies sample pretreatment for small-scale peptide N-glycosidase F (PNGase F) digests that use protein denaturation. Application Update 176: Preparation of Peptide N-Glycosidase F Digests for HPAE-PAD Analysis demonstrates that this new pretreatment method makes samples amenable to high-performance anion-exchange chromatography with pulsed amperometric detection (HPAE-PAD) oligosaccharide analysis with the CarboPac® PA200 column, thereby making it possible to monitor the oligosaccharide contentº of a glycoprotein. This approach overcomes the limitations of using traditional size-exclusion columns with low- to medium-molecular weight cut-off resins for sample pretreatment. Most size-exclusion methods are time consuming, require large amounts of sample, and run the risk of losing charged sugars.

The biotechnology industry has already produced a number of glycoprotein therapeutics and more are being investigated for their therapeutic potential.
